Available in the Life Sciences Building and funded by the Georgia Biotechnology Center of the Georgia Research Alliance, the High-Temperature Enzyme Purification Facility provides the equipment necessary for large-scale purification of proteins from thermophilic and hyperthermophilic microorganisms. The large-scale BioPilot Chromatography system was purchaed from Pharmacia Biotech. It consists of two computer-operated pumps which operate up to 100 mL/min together with a variety of pre-packed chromatography columns. Purification procedures can be monitored by UV-visible absorption, conductivity and/or pH. The system enables the large-scale purification of proteins in gram quantities by anion and cation exchange, gel filtration, hydroxyapatite, and hydrophobic interaction chromatography, using both medium- and low-pressure media. It allows facile scale-up from the conventional Phamacia FPLC system (maximum flow rate, 10 mL/min).
